The BOD5 test was established as a global standard following the work of the UK’s Royal Commission on Sewage Disposal between 1898 and 1915. They sought a reliable, repeatable method to quantify the polluting effect of sewage. The five-day incubation period was not chosen for purely scientific reasons but as a practical matter; it was determined that in UK rivers, water would typically reach the sea within five days, making this a relevant timeframe to assess the immediate oxygen-depleting impact. The 20°C temperature was selected as a representative average temperature for a surface river in the UK during summer. The test requires a sealed bottle, filled completely with the sample (or a diluted version) to prevent atmospheric oxygen from dissolving into the water during incubation. The sample is kept in the dark to inhibit photosynthesis, which would produce oxygen and interfere with the measurement of its consumption. The difference between the initial dissolved oxygen (DO) and the final DO after five days, adjusted for any dilution, gives the BOD5 value.
